Most hereditary disease articles ask you to watch for symptoms. This one does not, and the reason matters. JLPP has an early and narrow onset window. The OFA describes it as beginning “typically around 3 months of age,” and affected dogs do not reach adulthood. If your Black Russian Terrier is a healthy adult, it is not going to develop JLPP later.

What that means is that the entire value of DNA testing in this breed sits upstream — in the decisions breeders make before a litter exists, and in the questions buyers ask before they put down a deposit. There is no screening protocol, no early-intervention window, no diet or supplement that changes the outcome. There is only the mating decision.

That is actually the best kind of genetic problem to have. A recessive disease with a known mutation and a cheap, accurate test is a solved problem in principle. Whether it is solved in practice depends entirely on whether people test and share results.

What JLPP does — the clinical picture

The name of the disease describes its two halves. Laryngeal paralysis comes first: the nerves supplying the larynx degenerate, the vocal folds stop opening properly, and the puppy develops loud, raspy breathing on inspiration — worse when excited or exercising. Owners often notice that the bark has changed. Then the polyneuropathy becomes evident: weakness in the hind limbs, reduced or absent spinal reflexes, loss of sensation, low muscle tone, and an increasingly uncoordinated gait.

The companion paper on Rottweilers, Mhlanga-Mutangadura et al. (2016, Journal of Veterinary Internal Medicine 30(3):813-818), documented eight affected dogs and gives the clearest published tally of what the same mutation does:

  • Respiratory distress at 3-4 months in all eight dogs; laryngeal paralysis confirmed in seven.
  • Pelvic limb sensory ataxia and weakness in all eight.
  • Cerebellar ataxia in two, and megaesophagus in two — the latter explaining the regurgitation and aspiration risk.
  • Eye findings: cataracts in three, microphthalmia in two, persistent pupillary membranes in one.

Post-mortem, the defining feature is intracytoplasmic vacuolation within neurons — visible in the cerebellar cortex and cuneate nuclei of every dog examined, with loss of Purkinje cells leaving “empty baskets” behind. In one dog, the researchers traced the chain all the way out: marked nerve fibre loss in the recurrent laryngeal nerve, with matching muscle fibre loss in the cricoarytenoideus dorsalis — the muscle that opens the airway. The breathing problem is not in the lungs. It is in the nerve.

One mutation, two breeds — and what that tells us

The Black Russian Terrier was built in the mid-twentieth century from a small set of foundation dogs, and the Giant Schnauzer, Airedale Terrier and Rottweiler all contributed. So it is not a coincidence that the identical variant turns up in Rottweilers. Mhlanga-Mutangadura et al. (2016) genotyped affected Rottweilers and found all of them homozygous for c.743delC — the same deletion previously reported in Black Russian Terriers — and concluded that “the same founder mutation event was the source of the allele in both Rottweilers and BRTs.”

Their control data is worth quoting because it puts a number on the risk. Among 128 control Rottweilers, 105 carried two reference copies and 23 were heterozygous carriers — roughly 18% of that sample. Meanwhile, 468 dogs of other breeds were all clear. The mutation is real, it is common enough to matter, and it is confined to a small set of related breeds.

A third variant deserves a mention so you do not order the wrong test. Alaskan Huskies get a very similar syndrome from a different change in the same gene — a 218-base-pair SINE insertion in exon 7, reported by Wiedmer et al. (2016, G3 6(2):255-262). Same gene, different mutation, different assay. All three sit under OMIA:001970-9615. For a Black Russian Terrier you want the c.743delC test specifically.

The human parallel: Warburg Micro syndrome

RAB3GAP1 encodes part of a protein complex that regulates vesicle trafficking — the machinery cells use to move cargo around internally. It acts as a GTPase-activating protein for Rab3 and as a guanine nucleotide exchange factor for Rab18. When it fails, vesicles pile up, which is exactly what the neuronal vacuoles under the microscope represent.

In humans, loss-of-function variants in the same gene cause Warburg Micro syndrome 1 (WARBM1), characterised by microphthalmia, congenital cataract and neurological impairment. The overlap with the canine disease is striking — the eye findings in particular map closely. Wiedmer et al. noted that mice lacking the same gene have a much milder phenotype than either humans or dogs, which makes the affected dog a closer model of the human condition than the standard laboratory mouse. That is a real, if bittersweet, contribution from this breed to human medicine.

How to test in the United States, and what it costs

  4. Results come back in three categories: Normal (no mutation; can be bred to any result), Carrier (one copy; will never show signs, but passes it on half the time), and Affected/At-Risk (two copies).

The breeding rule, stated plainly

  • Clear × Clear — no carriers, no affected.

  • Carrier × Carrieron average 25% affected. This is the only combination that produces a sick puppy, and it is entirely avoidable.

The temptation, when a scary recessive disease is identified, is to remove every carrier from the gene pool at once. In a numerically small breed that reaction does real damage — it narrows the founder base and concentrates whatever else is segregating in the population. The BRTCA’s position, and the mainstream position in canine genetics generally, is to breed carriers to clears and select on everything else you care about: structure, temperament, hips, elbows, working ability.
